The DNA includes fragments ranging from 250-10,000 base pairs. The 1K and 3K bands have increased intensity to serve as reference points. The approximate mass of DNA in each band is provided (0.5 μg a load) for approximating the mass of DNA in comparably intense samples. This ladder is ready-to-use, and premixed with loading buffer dye for direct loading on gel.

Loading dye contains Bromophenol Blue as the tracking dye.

The BLUEstainTM 2 Protein ladder is a three-color protein standard with 13 prestained proteins, covering a wide range molecular weights from 3.5 to 245 kDa. Proteins are covalently coupled with a blue chromophore except for two reference bands (one green and one red band at 25 kDa and 75 kDa respectively) when separated on SDS-PAGE (Tris-glycine buffer).

The BLUEstainTM 2 Protein ladder is designed for monitoring protein separation during SDS-polyacrylamide gel electrophoresis, verification of Western transfer efficiency on membranes (PVDF, nylon, or nitrocellulose) and for approximating the size of proteins. The ladder is supplied in gel loading buffer and is ready to use. Do not heat, dilute or add reducing agent before loading.

Storage/Handling: Store at 4°C for 3 months. Store at -20°C for 24 months.

Description

ProGlowTM Western Blot Protein Ladders are designed for Western blots. ProGlowTM ladders can be used to monitor electrophoresis progress and to visualize protein standard bands directly on X-ray film or digital analytical equipment. ProGlowTM ladders consist of eight recombinant proteins in the range of 12-90 kDa. The 70 kDa protein is a prestained marker used for monitor of the electrophoresis process and evaluation of Western transfer efficiency. The other seven markers can bind to human, mouse, rat, goat, sheep and rabbit IgG with their IgG binding site, allowing direct visualization of markers on the X-ray film after ECL development.

The ProGlowTM Western Ladder will give varying signal intensity depending upon the concentration of your primary and secondary antibodies. GoldBio strongly recommends that you perform a test experiment with the ProGlowTM Ladder to determine the optimal amount to load on your gel. Run a test gel and Western blot in which 5 µl, 1 µl, 0.3 µl, and 0.1 µl of ProGlowTM Ladder is loaded, and examine the signal intensity.

Directions for Use:
1. Thaw to room temperature, and mix gently.
2. Load 1-5 µl ProGlowTM Western Blot Ladder. We recommend testing different loading amounts of the
    standards to determine the optimal amount to use under your experimental conditions.
3. After electrophoresis, transfer proteins to a NC or PVDF membrane.
4. Perform the blocking step followed by incubating with primary antibody and secondary antibody.
5. After the thorough washing, incubate membrane with ECL reagent, and then expose the X-ray film in
    the dark room.
6. ProGlowTM Western Blot Ladder is stable at 4°C for six month. Please put at -20°C for long term
    storage.
